DETERMINATION OF FAT CONTENT
SAMPLE PREPARATION
1.
Mix thoroughly the sample & place it in a mixer
grinder.
2.
Operate the grinder for 10 seconds.
3.
With the help of scoop mix well the content and operate
further for five seconds.
4.
Mix thoroughly and make it uniform mass on a butter
paper.
APPARATUS
1.
Soxhlet extraction apparatus.
2.
Air - oven. (0 - 105 °C).
3.
Glass - flat bottom flask - 250 ml
4.
Whatman Cellulose Extraction thimbles 25 mm x 80 mm.
5.
Desiccator
6.
Metler Electronic Balance (L.C - 0.1 mg.)
REAGENTS
Petroleum ether 60 /80 °C.
PROCEDURE
1.
Carefully weigh about 20 g of well-mixed powdered
sample in an extraction thimble.
2.
Plug the thimble with absorbent cotton, ensuring that
no material comes out.
3.
Place the thimble in the extractor and fit it to the
previously weighed flat bottom flask.
4.
Add about 200 ml of solvent to the flat bottom flask
(which contains glass beads). Affix the
condenser and allow through it a current of tap water to flow and heat the
flask.
5.
Continue the extraction for 6 hours.
6.
Remove the extractor from its bath, take out the
extraction thimble, and drain the solvent portion in the flask.
7.
Keep the Extraction thimble on the neck of the flask
for approximately 5 - 10 minutes so that complete draining of the solvent takes
place.
8.
Now remove the greater part of solvent in flask by
distillation from a hot - water bath.
9.
Heat the flask in an oven at 105 °C ± 2 ° C
for one hour to remove solvent.
10. Remove
the flask from oven and keep it in a desiccator to cool and weigh the flask.

Fat % = W2 - W1 x 100
W
W2 = Weight of the flask after
cooling
W1 = Weight of the flask before
extraction
W = Weight of the sample
